"Gain root remotely" is not a single plugin, but a family of them.

way to choose just that plugin through the command line

You can't enable plugins through the commandline. Instead, you specify a config file that lists which plugins are to be enabled / disabled. You could look into one of the third-party update-nessusrc scripts (Google is your friend) as a way to handle that, but that essentially requires a working config file to start with.

This scan would be taking place on the loopback address (127.0.0.1) of the Mandriva system.

Be aware that a few plugins are written such that they can not be run against the nessusd host.

Also, are there any tips to make the system be able to be picked up as gaining root remotely when the scan is run?

That depends on the specific plugin you need to demo. Study it, figure out what it checks for. And then configure your system so that the check succeeds.
